    • §  Xigris [drotrecogin alfa (activated)] revisions to the WARNINGS section of labeling based upon analyses of two clinical trial databases. Among patients with single organ dysfunction and recent surgery, all-cause mortality was numerically higher in the Xigris group compared to the placebo group. Patients with single organ dysfunction and recent surgery may not be at high risk of death and therefore may not be among the indicated population. Xigris should be used in these patients only after careful consideration of the risks and benefits.
    • Avonex (interferon beta-1a) revisions to the WARNINGS, PRECAUTIONS/Drug Interactions and ADVERSE REACTIONS/Post-Marketing Experience sections and Medication Guide. Severe hepatic injury, including cases of hepatic failure, has been reported in patients taking Avonex. The potential risk of Avonex used in combination with known hepatotoxic drugs or other products (e.g. alcohol) should be considered prior to Avonex administration.

  3. FDA Talk Paper: New Drug to Treat Type I and Type II Diabetes - Symlin, an injectable medicine to control blood sugar for adults with type 1 and type 2 diabetes. Symlin is to be used in addition to insulin therapy in patients who cannot achieve adequate control of their blood sugars on intensive insulin therapy alone.

    • Atacand (candesartan cilexitil), approved use in the treatment of chronic heart failure (CHF; New York Heart Association [NYHA] class II-IV and ejection fraction [EF] of 40% or less) to reduce the risk of death from cardiovascular causes and reduce heart failure–related hospitalizations as an alternative drug for patients who have developed tolerance to angiotensin-converting enzyme (ACE) inhibitor therapy, or it may be used in addition to ongoing therapeutic regimens (with or without ACE inhibitors).
    • Pegasys Plus Copegus (peginterferon alfa-2a [40 KD] and ribavirin) approved an expanded indication for combination therapy in the treatment of chronic hepatitis C virus (HCV) in patients coinfected with HIV.
    • Triage APAP TOX Screen (TOX Drug Screen with Acetaminophen) approved screening test the qualitative detection of acetaminophen in urine.
  2. FDA Alert:
    • Paxil CR (paroxetine hydrochloride) and Avandamet (rosiglitazone maleate + metformin hydrochloride) FDA and DOJ have seized the remaining stocks of Paxil CR and Avandamet tablets manufactured by GlaxoSmithKline, Inc. Manufacturing practices failed to meet the standards laid out by FDA.
    • Elidel (pimecrolimus) and Protopic (tacrolimus) Public health advisory to inform about a potential cancer risk from use based on information from animal studies, case reports in a small number of patients, and how these drugs work.

    • Abilify (aripiprazole) expanded indication for use in bipolar I disorder after a recent manic or mixed episode who have been stabilized and maintained for at least six weeks.
    • Isovorin (intravenous levofolinic acid) two orphan drug designation allowing use with 5-fluorouracil for the treatment of colon cancer, and in conjunction with methotrexate (MTX) for osteosarcoma.

    • Vistaril (Hydroxyzine Pamoate) use is contraindicated in patients who have demonstrated prior hypersensitivity to the active moiety or components of the products. Inert components of the capsule include hard gelatin (may contain Yellow 10, Green 3, Yellow 6, Red 33, and other dyes), magnesium stearate, sodium lauryl sulfate, starch, and sucrose. Inert components of the oral solution include carboxymethylcellulose sodium, lemon flavor, propylene glycol, sorbic acid, and sorbitol solution.
    • Estraderm (Estradiol Transdermal System) revised contraindications and warnings associated with its use. Use of estrogens, including the estradiol patch, is contraindicated in individuals with liver dysfunction or disease. The FDA also warned of the increased risk of developing probable dementia associated with the use of conjugated estrogen (CE) plus medroxyprogesterone acetate (MPA) therapy compared with placebo in postmenopausal women aged 65 years and older. The FDA notes that it is unknown whether the warning applies to younger postmenopausal women or to those taking estrogen-only therapy.Results of the WHI also showed increased risks of myocardial infarction, stroke, invasive breast cancer, pulmonary emboli, and deep vein thrombosis in postmenopausal women (aged 50-79 years) during five years of treatment with CE plus MPA.
    • Nolvadex (Tamoxifen) revised safety labeling to warn of the risk of uterine malignancies that may be associated with its use based on results of the Breast Cancer Prevention Trial that demonstrated increased risks of endometrial adenocarcinoma and uterine sarcoma in patients receiving tamoxifen compared with placebo (incidence rates per 1,000 women-years, 2.20 vs 0.71 and 0.17 vs 0.04, respectively).

    • Regenafil, demineralized bone matrix (DBM) allograft paste products for use in spinal and orthopaedic applications to fill voids in bone. The products consist of allograft DBM and a gelatin carrier. The DBM products are packaged and marketed under different names according to their application: Regenafil and Regenafil RT allograft pastes are used to fill small gaps in bones of the maxillofacial area; Optefil and Optefil RT allograft pastes/syringes are used to fill gaps in bone; Osteofil and Osteofil RT DBM pastes are used as bone void fillers in cervical and lumbar procedures.

  1. §  MEDWATCH: Tysabri (natalizumab) Marketing of Tysabri (natalizumab) has been suspended due to two serious adverse events reported with its use. FDA received reports of one confirmed, fatal case and one possible case of progressive multifocal leukoencephalopathy (PML) in patients receiving Tysabri for multiple sclerosis. Although the relationship between Tysabri and PML is not known at this time, because of the rare, serious and often fatal nature of PML, FDA announced the following, effective immediately:
    • Biogen Idec is voluntarily suspending marketing of Tysabri.
    • Biogen Idec is suspending dosing of Tysabri in clinical trials and is notifying patients and investigators of the possible association between Tysabri and PML.
    • Patients being treated with Tysabri should contact their physician to discuss appropriate alternative treatments.
